Lung cancer is one of the most common cancer and the leading cause of death in the world and our country. According to the 2020’s data of the World Health Organization, lung cancer is the second one of the most common cancers in Turkey. At the same time, it is the first one of the most common cancer in men in Turkey. The five year survival rate is 20.5% because lung cancer metastasis very quickly and early diagnosis is difficult. Lung cancer consists two main groups as small cells lung cancer (SCLC) and non-small cell lung cancer (NSCLC). NSCLC more common than the SCLC. NSCLC is divided tree subtypes. These are adenocarcinoma, squamous cell carcinoma and large cell carcinoma. Stem cells are undifferentiated cells with unlimited division and self-renewal ability. According to the cancer biology, there are 3 options between stem cells and cancer cells relationship. One of them is that, tumors contain cancer stem cells (CSC). CSC were first documented for leukemia and multiple myeloma. After than it also observed in solid tumors. Cancer stem cells are responsible for the initiation of the tumor. CSC have negative effects on metastasis and drug resistance and its increase cancer’s incidence and mortality rate. In lung cancer, cancer stem cells are assumed to derive from more than one source. So, they should be investigated for treatment. These studies are carried out with cancer stem cells isolation and characterization. Although a universal marker for cancer stem cell isolation has not been identified, brain, hematopoietic, prostate, and colon cancer stem cells display expression of the membrane antigen CD133, which is shared by normal stem cells. CD133 positive cells show characteristics of lung cancer stem cells and thus CD133 is an important marker of lung cancer stem cells. For this reason, in this study, CD133 (+) lung cancer stem cell isolation and characterization was performed by MACS method from the H460 cell line, which is a sub-member of non-small cell lung cancer and a large cell carcinoma model.
